SNP Discovery & Validation

As the most common form of genetic variations in most organisms, SNPs are a major focus in studies of discovery, genotyping and genetic analysis of genome polymorphisms. SinoGenoMax offers complete custom panel genotyping from sample preparation to final analysis for various types of SNP assays. We have successfully aided our customers in SNP discovery and genotyping of more than 300 human genes related to common diseases, such as hepatitis B, cancer, hypertension, and coronary heart disease, etc.

SNP discovery and validation based on direct PCR re-sequencing
We help the customers in experimental design and PCR optimization. All PCR reactions are set up in 96 or 384 well plates. PCR products are purified and processed for the subsequent sequencing reactions. All fragments are sequenced in both forward and reverse directions to ensure maximum reliability of the results. The traces are then transferred to our proprietary software for automated SNP detection. Extensive quality control procedures (both manual and computational) help us further refine the quality of data.

SNP genotyping based on TaqMan technology
SinoGenoMax offers both custom-designed and off-the-shelf ABI TaqMan SNP genotyping via ABI PRISM 7000 Sequence Detection System. Both SNPs and insertion-deletions of up to six bases can be detected.

SNP genotyping based on SNPLex technology
The SNPlex system from ABI uses Oligonucleotide Ligation Assay (OLA) and PCR technology for allelic discrimination and ligation product amplification. The genotype information is then encoded into a universal set of fluorescence-labeled, mobility modified fragments for rapid detection by capillary electrophoresis on ABI 3730xl DNA analyzers. SNPlex allows multiplexing reactions up to 48 different SNPs per reaction; therefore one 3730xl is capable of performing over 200,000 SNP genotypes per day, lowering the cost of SNP analysis. SinoGenoMax will help the customer with design and ordering of locus specific oligos from AppliedBiosystems, and will conduct the assays starting from sample preparation until data analysis and report.
SinoGenoMax is the only SNPLex technology provider in China which authorized by ABI Company.Our track record includes successful completion of a large scale genotyping project for an overseas research institute in which over 3,000 samples were genotyped for ~ 300 SNP markers.

DHPLC
DNA sample pooling technique allows the identification of homozygotic mutation as well as a heterozygotic type, which improves the SNPs discovery efficiency.
