Functional Genomics and Drug Discovery Platform
Genome-Based Target Validation
SinoGenoMax has assembled a genome-based drug target validation platform to systematically study human gene functions and to discover and develop proprietary genome-based drugs. The platform consists of following systems:
♦ Data mining system for human genes
♦ High-throughput expression system for recombinant human proteins
♦ High-throughput and high-content functional screening system for human genes
Using this platform, SinoGenoMax has analyzed nearly 4000 human gene sequences, cloned and expressed about 500 genes with more than 85% of which are novel genes. These genes have been registered and annotated in the in-house database. This system has built a solid foundation for genome-based target validation.
Pathway-Based Target Validation
Our pathway-based target validation platform has been developed to screen unknown functions of the kinase genes:
♦ Reporter assay
♦ Yeast two hybrid system
♦ siRNA screening assay
♦ GeneChip screening assay
We have developed reporter assays for over ten key signaling pathways, through which we had carried out functional screens over five hundred times. Several protein kinases which play key roles in the disease signaling pathways such as cancer cell division and immune suppression have been identified.
siRNA-Based Drug Screening
SinoGenoMax has developed proprietary multiple transient and stable dual-promoter siRNA expression vectors and a rapid siRNA validation system. By taking advantage of DNA chip technology with our siRNA vector system, we now make the high-throughput validation of siRNA possible.
SinoGenoMax has also constructed a random siRNA library, which is being used to systematic screen for gene function. Through extensive international collaborations, SinoGenoMax is developing siRNA plasmid libraries which cover the whole human and mouse genome to facilitate siRNA drug screening and development. |
